Four layers, one single reading

    Microbiome

    Metagenomic NGS/Nanopore: who is there and in what proportion, at species resolution.

    Functional metabolomics (SCFA such as butyrate): what the ecosystem produces and how it feeds the barrier.

    Permeability and inflammation

    Fecal calprotectin (a validated inflammation marker) and zonulin as an accessory barrier datum.

    Intolerances

    Breath tests (SIBO with hydrogen and methane; fructose) to complete the symptom map.

Clinical rigour: we report each biomarker according to its level of evidence. Fecal calprotectin is a validated inflammation marker; zonulin is offered as an accessory permeability datum, whose immunoassay is subject to scientific debate, and never as a standalone diagnosis of «leaky gut». Sophia contextualises each datum and recommends correlating it with the clinical picture and with validated markers.
